Professor of Biochemistry & Director, Center in Molecular Toxicology Vanderbilt University School of Medicine On "Recent Thoughts about Cytochrome P450: Ligand Specificity & the use of P450s in Drug Discovery "Abstract Cytochrome P450 (P450) enzymes are remarkably diverse oxygenation catalysts and are found throughout nature. Understanding their selectivity is challenging, and many efforts have been made in the context of predicting interactions with ligands. Defining the selectivity of a P450 in terms of a single residue may be difficult due to the global nature of proteins and their functions. A recent example comes from site-directed mutagenesis and other work with P450 2D6 and the roles of the acidic residues at positions 301 and 216. Although most of the interest regarding P450s in the pharmaceutical industry has been in the drug development aspects, P450s also offer potential in discovery, not only of drugs but also other useful chemicals. Potential applications range from use of P450s as drug targets to the use of randomly-generated mutants of P450s to produce libraries of new chemicals and drugs. ___________________________________________________________________________
